摘要
采用大鼠黄体细胞体外培养的方法,观察了心房钠尿肽(ANP)对黄体细胞孕酮生成的影响及与细胞内信使cAMP的相互关系。发现ANP能抑制黄体细胞孕酮的基础分泌及人绒毛膜促性腺激素(HCG)刺激下的孕酮分泌。当浓度10-7mol/L的ANP加入培养的黄体细胞悬液30min后,即发生明显地抑制孕酮分泌作用(P<0.01);腺苷酸环化酶激动剂Forskolin则可促进黄体细胞分泌孕酮;当Forskolin与ANP共同作用时,ANP可抑制Forskolin对黄体细胞的促进作用,孕酮分泌恢复到对照组水平。实验结果提示:ANP可能是一种卵巢局部调节肽,它可能通过抑制小黄体细胞内的腺苷酸环化酶,减少CAMP的生成和释放,进而抑制小黄体细胞的孕酮分泌。
The effect of ANP on progesterone production and the relationship between ANP and intracellular messenger CAMP were investigated by incubated rat luteal cells in vitro.It was found that ANP could inhibit the basic progesterone production and progesterone secretion induced by HCG from rat luteal cells.When ANP was added at a concentration of 10 -7 mol/L into suspension of luteal cells for 30 min,the progesterone production decreased markedly( P <0 01).Forskolin,an excitant of adenylcyclase could promote the progesterone secretion,but inhibited by ANP.This study suggests that ANP might be a local-ovarian regulatory peptide which inhibits progesterone secretion from rat small luteal cells through inhibiting the activity of adenylclase and decreasing CAMP accumulation.
