摘要
目的 观察睫状神经生长因子 ( ciliary neurotrophicfactor,CNTF)和雪旺细胞神经营养活性物质 ( Schwann cellsneurotrophic agent,SCNA)对视神经损伤后轴突的形态和纤维数目变化的作用 .方法 采用镊夹法制作大鼠视神经损伤模型 ,损伤后立即一次性向玻璃体内注射 CNTF( 5 0 ng)或SCNA ( 10μL ) ,在损伤后 1,2 ,3和 4 wk时观察夹伤视神经的形态 ,及进行图像分析和轴突纤维计数 .结果 损伤后视神经轴突减少 ,间质增多 .CNTF组和 SCNA组的轴突数密度在 1wk时分别为正常值 ( 0 .10 65± 0 .0 0 5 6个·μm- 2 )的0 .5 9和 0 .61,而对照组为 0 .5 5 ,实验组与对照组比较差异显著 ( P<0 .0 5 ) .但在夹伤 2 wk后 3个组间、各时间点间数据差异均不显著 ( P>0 .0 5 ) .结论 CNTF及 SCNA一次玻璃体给药能在 1wk内减缓视神经损伤后轴突数目的减少 .
AIM To study the effects of ciliary neurotrophic factor (CNTF) and Schwann cell neurotrophic agent (SCNA) on changes of configuration and the number of nerve fibers ina rat's model of the crushed optic nerve. METHODS The optic nerve injury model of rats was induced by crushed at the optic nerve 1 mm retrobulbarlly. Then an intravitreal injection of CNTF (50 ng) or SCNA (10 μL) was performed. The cross section morphology changes were observed 1, 2, 3 and 4 weeks after injury, and the number of nerve fibers was determined by using a computerized image analysis system at the time points. RESULTS The axon number of crushed optic nerve decreased but the matrix increased. The axon number density of the CNTF group and the SCNA group was 0.59 and 0.61 of the normal level (0.1065±0.0056) one week after injury, while the control group's was 0.55 ( P <0.05). But there was no difference in three groups 2, 3 and 4 weeks after injury. CONCLUSION An intravitreal injection of CNTF or SCNA can reduce the loss of optic nerve fibers in crushed optic nerve, so far only noted within one week after injury.
