摘要 目的 为论证“方剂体内 /血清成分谱”概念 ,建立同步测定人血清中阿魏酸、川芎嗪的HPLC法。方法 以香豆素为内标 ,甲醇 水 醋酸 (40∶5 9.7∶0 .3 ,v/v)为流动相 ,InertsilODS 3色谱柱 (15 0mm× 4.6mm ,5 μm)为固定相 ,流速 1.0mL/min。以二极管阵列检测器同时获取 32 2nm和 2 79nm不同波长下的数据。结果 阿魏酸、川芎嗪和内标分离完全 ,阿魏酸在 (40 .16~ 80 32 )ng/mL范围内线性关系良好 (r=0 .9978) ,变异系数≤ 10 % ,平均回收率为 10 0 .3 % ,最低检测限为2 .5 1ng ,最低检测浓度为 2 5 .1ng/mL ;川芎嗪在 (39.4~ 7880 )ng/mL范围内线性关系良好 (r =0 .990 6 ) ,变异系数≤10 % ,平均回收率为 96 .37% ,最低检测限为 1.97ng ,最低检测浓度为 2 3.6 4ng/mL。 OBJECTIVE To demonstrate the concept of 'in vivo/serum component spectrum of Chinese drug' and establish the simultaneous determination method of ferulic acid and ligustrazine with HPLC. METHOD The experiment was conducted with coumarin as the interior label, methanol water acetic acid (40∶59.7∶0.3, v/v) as the mobile phase and Inertsil ODS 3chromatigram column (150 mm×4.6 mm, 5 μm) as the fixed phase, at the rate of 1.0 mL/min. Numerical date at the wavelengths of 322 nm and 279 nm were obtained with a diode array tester. RESULT The isolation of ferulic acid and ligustrazine was complete with good linear relation of ferulic acid at the range of (40.16~8 032)ng/mL ( r =0.997 8), a variation coefficient≤10%, an average recovery rate of 100.3%, the lowest determination limit of 2.51 ng and the lowest determination concentration of 25.1 ng/mL; and with a good linear relation of ligustrazine at the range of(39.4~7 880)ng/mL ( r =0.990 6), a variation coefficient≤10%, an average recovery rate of 96.37%, the lowest determination limit of 1.97 ng and the lowest determination concentration of 23.64 ng/mL. CONCLUSION This method can be used to determine ferulic acid and ligustrazine in human serum.
