胃癌组织中CASP-3和bcl-xl表达的研究

Expression of Caspase-3 and bcl-xl in Gastric Carcinoma

目的 探讨胱氨酸蛋白酶 3(CASP 3)和bcl xl表达与胃癌预后的关系 ,及CASP 3与bcl xl表达相互间的关系。方法 应用免疫组织化学ABC法检测人胃癌组织中CASP 3和bcl xl的表达情况。结果 正常胃粘膜上皮和胃癌组织中CASP 3的阳性率分别为 96 .0 % ( 4 8/ 5 0 )、70 .0 % ( 35 / 5 0 ) ,两者比较有显著性差异 (P <0 .0 5 ) ;正常胃粘膜上皮和胃癌组织中bcl xl的阳性率分别为 42 .0 % ( 2 1/ 5 0 )、6 6 .0 % ( 33/ 5 0 ) ,两者比较有显著性差异 (P <0 .0 5 )。CASP 3和bcl xl的表达与胃癌的预后指标 (如组织学类型 ,TNM分期等 )明显相关 (P <0 .0 5 ) ,但CASP 3与bcl xl表达之间无明显相关性 (P >0 .0 5 )。结论 人胃癌组织中CASP 3的表达较低而bcl xl的表达较高。CASP 3低表达和bcl xl的高表达导致的细胞凋亡异常降低及增生过度 ,可能是胃癌的发病原因之一。CASP 3与胃正常粘膜及肿瘤细胞的凋亡有关。CASP 3和bcl

Objective To study the relationship between the expression of caspase 3/bcl xl and the prognostic indexes in human gastric adenocarcinoma.and the relationship between expressions of caspase 3 and bcl xl.Methods The expressions of caspase 3 and bcl xl in 50 specimens of human gastric adenocarcinoma were studied by immunohistochemistry.Results The caspase 3 expression was detectable in 70.0%(35/50) cancer specimens and in 96.0%(48/50) of normal tissue( P <0.01).The bcl xl expression was dectectable in 66.0%(33/50) and 42.0(21/50)( P <0.05).Caspase 3 significantly correlated to the prognostic index,such as pathologic type and TNM staging.So does bcl xl.But there is was no significant correlation between the expressions of caspase 3 and bcl xl.Conclusion There are low expression of caspase 3 and high expression of bcl xl in human gastric adenocarcinoma.The gastric adenocarcinoma is associated with an inhibition of apoptosis and the augmentation of proliferative activity.These results also suggest that caspase 3 is involved in the development or regulaion of apoptotic cell death in normal and neoplastic mucosa of the human stomach.Caspase 3 and bcl xl may be are two useful prognostic indexes.
