摘要
目的 :探讨刺激性三磷酸鸟苷结合蛋白α亚单位 (Gsα)基因限制性内切酶 (FokI)多态性与国人原发性高血压及其左心室结构之间的关系。 方法 :采用多聚酶链反应限制性片断长度多态性 (PCR RFLP)方法检测 78例健康人 (正常对照组 )和 14 2例原发性高血压患者 (高血压组 )Gsα基因FokI多态性 ,超声测定患者的左心室结构。 结果 :①高血压组Gsα基因FokI等位基因频率与正常对照组比较有显著性差异 (P =0 0 18) ;②高血压组不同基因型患者间的左心室后壁厚度有显著性差异 (P =0 0 40 )。 结论 :提示Gsα基因FokI多态性与国人原发性高血压及其左心室后壁厚度相关。Gsα基因可能是原发性高血压的遗传因素。
Objective:To investigate whether the gene of the G s protein a subunit(G sα) is implicated in genetic factors of essential hypertension (EH) in Chinese population. Methods:The polymorphism of G sα gene was detected by polymerase chain reaction combined with restriction enzyme digestion in 142 hypertensive patients and 78 normotensive controls.Parameters of left ventricular structure were assessed by echocardiographic method in 99 patients. Results:There was significant difference in frequency of the FokI alleles between patients(FokI+∶FokI- 40.8%:59.2%)and controls(FokI+∶FokI- 29.5%∶70.5%)(p=0.018).The posterior wall thickness differed significantly among FokI--(9.4±1.3*!mm),FokI-+(9.3±1.2*!mm),FokI++(8.5±1.1*!mm)genotypes in hypertensive patients(p=0.040). Conclusion:The FokI polymorphism of G sα gene is associated with EH and their posterior wall thickening in chinese people.These suggest that a functional trait locus in or near the G sα gene may contribute to EH.
