摘要
【目的】探讨玻璃体内插入外周神经和玻璃体内注射 3 异丁基 1 甲基黄嘌呤 (IBMX)对轴突损伤的视网膜节细胞再生的影响。【方法】 2 0只成年金黄地鼠随机分 4组 ,每组 5只动物。切断视神经 (ON)并缝接坐骨神经 (attachedgraft ,AG)为再生对照组 ;ON切断并缝接坐骨神经后再于玻璃体内注射IBMX和 /或插入一小段自体坐骨神经 (SN)为实验组。采用逆行示踪标记术 ,观察视网膜平铺片节细胞数。【结果】①对照组 (AG)每个视网膜再生的节细胞数为 (142 8± 2 84)个 /reti na;②外周神经组 (AG +SN)为 (2 2 2 0± 415 )个 /retina ;③IBMX组 (AG +IBMX)为 (2 42 4± 10 2 6 )个 /retina;④外周神经和IB MX联合组 (AG +IBMX +SN)为 (30 6 5± 6 5 4)个 /retina;其中 ,AG +SN组同AG组相比存在显著性差异 (P <0 0 1) ;AG +IBMX +SN组同AG、AG +IBMX和AG +SN组相比均有显著性差异 (P <0 0 1)。
To investigate the effects of transplantation of sciatic nerve(SN) and injection of 3 isobutyl 1 methylxanthine(IBMX) into the vitreous on axotomized retinal ganglion cells (RGC) in adult hamsters. 20 adult golden hamsters were ramdomly divided into 4 groups. Optic nerve (ON) was transected and a segment of autologous sciatic nerve (attached graft, AG) was removed and sutured to the proximal stump of ON in regenerating control group (AG group). Animals in the experimental groups were further treated with IBMX injection and/or implantation of a short of segment sciatic nerve (SN) intravitrously. Use the fluorescent retrograde labeling method to measure the quantity of RGC. ①In control (AG) retinas, the mean number of regenerating RGC was (1 428±284)/retina, ②In AG+SN group, the mean number was (2 220 ±415)/retina, ③In AG+IBMX group, the mean number was (2 424±1 026)/retina, ④The mean number in AG+IBMX+SN group was (3 065±654)/retina. AG+SN group was significantly different from AG group (P<0 01). AG+IBMX+SN group was significantly different from AG, AG+IBMX and AG+SN groups (P<0 01). [Conclusions] The results indicate that SN combined with IBMX exert significant effects on promoting the regeneration of RGC after axotomy.
