摘要
目的 研究骨髓瘤细胞系KM3中免疫球蛋白重链基因 (IgH)的易位。 方法 用PCR方法合成地高辛标记的各IgH转换区 (switchregion ,S区 )探针 ,即σμ/σδ、Sμ、Sγ、Sα、Sε的 3′端和 5′端探针 ,应用Southernblot方法检测杂交结果。结果 胎盘基因组DNA中有一 10 .0kb的HindⅢ片段可同时与 5′Sμ探针和 3′Sμ探针杂交 ,而骨髓瘤细胞系KM3基因组DNA中除有一相同的 10 .0kb的HindⅢ片段外 ,还有 1个 5 .6kb的HindⅢ片段 ,仅与 3′Sμ探针杂交。 结论 骨髓瘤细胞系KM3存在 1个由IgH基因易位引起的异常重组片段 。
Objective To identify the IgH translocation in KM3 myeloma cell line established from a Chinese patient. Methods Southern blotting was performed with dig labeled probes specific for IgH switch regions to identify and distinguish IgH switch recombination events. Results Besides a 10.0 kb Hind Ⅲ fragment being able to hybridize with both 5′Sμ and 3′Sμ probes as that in placenta genome DNA. There was a 5.6 kb Hind Ⅲ fragment in KM3 genome DNA could only hybridize with the 3′Sμ probe. Conclusion There was an illegitimate switch recombination fragment containing 3′Sμ region in the KM3 myeloma cell line, which was caused by the IgH translocation involving the IgH Sμ region.
