摘要
目的 :探讨针刺对快速老化模型小鼠 (SAM- P/10 )肝抗氧化酶活性及基因表达的影响。方法 :采用生化及分子生物学技术 Northern Blot方法 ,观察针刺水沟、内关、太冲穴后 ,SAM- P/10小鼠肝丙二醛含量 (MDA)、超氧化物歧化酶 (SOD)活性及其基因表达水平的改变。结果 :针刺可降低模型动物自由基代谢产物 MDA含量 ,提高抗氧化酶活性 ,并对 SOD m RNA表达有上调作用 ,且具有腧穴的特异作用。结论 :针刺可在基因调控水平影响抗氧化酶活性 ,减少自由基代谢产物 。
Objective:To study the effects of acupuncture on the activ ity of antioxidase and expression o f gene in liver of SAM-P/10Method s:DNA content,SOD activity and SOD mRNA levels were assayed by bioch emical and Northern Blot methodsR esults:The level of MDA in liver of the acupoint group was lower than that i n the control group The higher defense defense capacity against oxygen free radi cal was supported by the findings that the activitiy of SOD was hig her than that in the control groupThe SOD mRNA level demonstrated the s ame tendency as the enzyme changes Conclusion:Acupuncture can affect the activity of antioxidase,the c apacity to remove oxygen free radi cal and up-regulation of SOD mRNA at the transcription level
