摘要
目的:探讨结肠直肠癌组织基质金属蛋白酶-2(matrixmetalloproteinase2,MMP2)和基质金属蛋白酶-9(matrixmetalloproteinase9,MMP9)的表达与肿瘤血管形成的关系。方法:应用免疫组化LSAB法进行研究,抗人MMP2、抗人MMP9和抗人CD31单克隆抗体分别用于检测72例结肠直肠癌手术切除标本MMP2、MMP9的表达情况及肿瘤微血管密度(microvesseldensity,MVD)。结果:72例结肠直肠癌组织中MMP2、MMP9的表达阳性率分别为69.4%和73.6%,DukesA、B、C、D各期的MMP2、MMP9的表达阳性率有递增的趋势(P值分别为0.013和0.027)。肿瘤血管形成与Dukes病理分期也存在一定联系,预后较好的DukesA、B期的MVD值明显低于预后较差的DukesC、D各期(P值<0.001)。MMP2、MMP9的表达情况与肿瘤血管形成的程度显著相关,MMP2、MMP9的表达阳性者的MVD值明显高于MMP2、MMP9的表达阴性者(P值分别为0.033和0.043)。结论:MMP2、MMP9在结肠直肠癌的血管形成中起重要作用,MMP2、MMP9可通过促进肿瘤血管形成影响肿瘤的进展。
Objective:To investigate the expression of m atrix metalloproteinase-2(MMP-2)and matrix metalloproteinase-9(MMP-9 )in colorectal cancer and their relationship with angiogenesis.Methods :The e xpression of MMP-2and MMP-9in 72sections of colo rectal adenocarcinoma and microvessel density(MVD)in the neoplasms were determined by LSAB technique of immu nohistochemical staining with monoclonal antibodies against MMP-2,MM P -9,and CD31.Results:The positive rates of the expression of MMP-2and MM P-9in 72sections were69.4%and 73.6%,respectively.The positive ra tes of MMP-2and MMP-9were increased according to Duke' s stage(P values wer e 0.013and0.027,respectively).Angiogenesis was also related with Du kes stage.MVD was significant hi gher in later stage than in earlier stage(P <0.001).Both the expression of MMP-2and MMP -9were correlated with a ngiogenesis.MVD in MMP-2or MMP-9positive samples was higher than those with negative expression(P values were 0.033and 0.043,respectively).Co nclusions :Both MMP-2and MMP-9are important fo r angiogenesis in colorectal adenocarcinoma.MMP-2and MMP-9may affect the progression of tumor b y promo tion of angiogenesis.
