棉胚珠继代培养纤维体系的建立(英文)

A System of Ovule Subculture for Cotton ( Gossypium hirsutum L.) Fiber Develo pment

针对传统的棉胚珠培养方法因纤维在发育中途夭亡而不能用于研究次生壁增厚和强度形成的机理 ,本文建立了棉胚珠继代培养纤维体系 ,即根据花铃的形态特征选取 类发育状态的胚珠作外植体 ,以 14d为周期连续 3次继代 ,在继代培养基中添加 5 g· L-1活性碳 ,并根据纤维发育不同阶段的特点调整植物生长物质的种类和浓度。结果使纤维重现了植株条件下的发育历程 ,其比强度、长度和细胞壁厚度等品质指标达到或接近植株条件下发育的纤维 。

To overcome the limitations of the method of cotton ovule culture deve loped by Beasley and Ting, a system of ovule subculture for cotton fiber develop ment (SOSCFD) has been established, including the following main points: The ovu les in 2nd developmental state determined by the characteristics of corolla and stigma were used as explant. Cultured cotton ovules were successively transferre d to fresh media with 14 cyclic days each subculture. In subculture media activa ted charcoal (5 g·L -1 ) was supplemented and the constituent of plant grow th regulators and its concentration were changed to accord with the demands of f iber develo pment in different phases. The specific strength, cell wall thickness and length of fiber grown in SOSCFD, exhibiting a developmental schedule similar to fiber grown in plant, reached or approached those grown in plant. Therefore, SOSCFD ca n be used as a new experimental tool of studying the characteristics of each sta ge and whole process of fiber development, especially the formation of secondary cell wall thickness and strength of fibers.
