摘要 本研究对从中间偃麦草与小麦杂种后代中选育的小偃麦种质系 GP143,在进行白粉病和条锈病抗性鉴定的基础上 ,对其进行了细胞学和 RAPD鉴定。结果证明 GP143兼抗白粉病和条锈病 ,其根尖细胞染色体数为 4 2 ,花粉母细胞减数分裂中期 I(PMC MI)染色体构型为 2 n=2 1 ,在它与小麦的杂种 F1PMC MI染色体构型中 ,常观察到四价体 ;对 GP143及其双亲进行 RAPD分析 ,从 4 0个随机引物中筛选出 4个引物能够扩增出中间偃麦草亲本的特异 DNA片段。初步确定 Tritelytrigia germplasm line GP143 with good resistance to powdery mildew and stripe rust, which came from the progeny of E l ytrigia intermedium and Yannong15, was identified by cytology and RAPD analysi s. The results showed that chromoso me number of GP143 in root tip cell was 42, chromosome configuration of GP143 in PMC MI was 2n=21Ⅱ, and quadrivalent was observed in PMC of (F 1) hybrid betwe en GP143 and different wheat varieties. GP143 and its parents were further analyzed by RAPD method. 40 random primers were used for RAPD amplifi cation, the speci fic DNA of E.intermedium can be amplified by 4 of them in GP143. Therefore, GP143 was proved as a translocation line.
