摘要
利用反转录 (RT)及套式PCR(N -PCR)扩增并测定了 6株具有不同表症近期甘肃省猪瘟流行野毒及C -株细胞疫苗毒的主要免疫原E2基因的核苷酸序列。序列分析比较结果表明 ,6株流行野毒与C -株疫苗毒的核苷酸同源性为 82 %~ 84 % ,流行野毒之间的核苷酸同源性在 89%~99%之间 ,并且明显可分为二个组群 ,病毒株所属组群与其临床症状有一定的相关性。流行野毒与C -株毒在中和抗原决定簇上有部分氨基酸存在性质差异 ,可能影响C -株毒对流行野毒的中和滴度。
The major immunogen E2 gene of the cell-adapted Chinese hog cholera virus(C strain cell vaccine virus) and six strains of recent prevalent field virus with different symptoms that were collected from Gansu province,were amplified by reverse transcription(RT) and nested-polymerase chain reaction(N-PCR),and their nucleotide sequences were determined.The results of sequence analysis and comparison showed that,the homologies of the prevalent field viruses with C-strain cell vaccine virus are about 82%~84%,the prevalent viruses with each other about 89%-99%,they can apparently divided into two groups,there are certain relativity between the groups that the virulent strains belong to and their clinical symptoms.In their neutral cluster,there are differences in amino acid property between the prevalent field viruses and C-strain cell vaccine virus,these may have effect on neutral titer of C-strain virus to the prevalent field virus.
