摘要
目的 探讨去势大鼠骨质变化规律及三苯氧胺对去势大鼠骨质疏松骨量变化的影响。方法 选用健康 2月龄雌性SD大鼠 30只 ,随机分为虚假手术组、卵巢切除组 (去势组 )和三苯氧胺治疗组 ,所有动物手术后第 11周和第 12周先后二次行腹腔注射茜素红以作活体荧光标记 ,第二次标记后第二天处死所有动物 ,取尾椎近节制作不脱钙骨切片进行骨组织形态学的观察 ,了解骨量变化情况。结果 去势组骨量明显减少 ,骨吸收增强 ,成骨作用减弱 ,三苯氧胺组骨量接近正常。
Objective To sudty the developmental regulation of osteoporosis in oveariectomized rats and the antagonism of tamoxifen to bone mass alteration in estrogen deficient condition. Methods\ Post\|menopausal osteoporosis rat model was designed by bilateral ovariectomy. Thirty 2\|month\|old female SD rats were randomly divided into three groups:sham\|operated, bilaterally ovariectomized, and treated with tamoxifen. The effects of 12 weeks' treatment with tamoxifen on trabecular bone mass of vertebrae were studied histomorphometrically. Alizarin was injected i.p. to each rat on two separate occasions to label the sites of bone formation. All rats were sacrificed two days after the second injection. Two proximal vertebrae of the tail were processed and sectioned undercalcifiedly for static and dynamic quantiative bone histomorphometric study, in order to observe bone mass alteration. Results\ The skeletal alteration in the ovariectomized rats was obvious bone loss with decreased bone formation and increased bone resorption. The bone mass in the tamoxifen treated group was close to normal. Conclusion\ Tamoxifen can effectively antagonize the bone wass alteration caused by ovarian hormone deficiency.
