小鼠皮肤癌变过程中GST-π免疫组化染色观察 被引量:8

摘要 为探讨谷胱甘肽s-转移酶(GSTs)在小鼠 皮肤癌变中的作用,本实验利用GST-π抗体和免疫组织化学方法,观察GST-π在小鼠正常皮肤(n=6)、良性增生(n=8)、异性增生(n=12)、鳞状细胞癌(n=9)中的表达和作用部位。结果表明,在正常皮肤标本中GST-π几乎没有表达;在良性增生中,细胞质染色为50%,细胞核周染色为12.5%;在异性增生中细胞质染色为83.3%,细胞核周染色为75%;在鳞状细胞癌细胞质染色为100%,细胞核周染色为88.9%。GST-π在小鼠皮肤癌变过程中含量明显增高,尤其GST-π在皮肤角化层和核周的表达,可以作为诊断癌症的一种标志。 The expression and localization of glutathione s-transferase n isoenzyme in mouse normal skin(n = 6), hyperplasia(n = 8), dysplasia( n = 12) ,and squamous cell carcinoma(n= 9) of mouse skin lesions were examined immunohistochemically using polyclonal antibody raised against GST-π with the standard peroxidase anti-peroxidase(PAP)method. GST-π was almost absent in the epithelium of normal mouse skin and overlying benign fibrous tissues. GST-Tr was expressed in 50% cytoplasmic staining and 12. 5% peri-nuclear staining in hyperplasia,83. 3% cytoplasmic staining and 75% peri-nuclear staining in dys-plasia, 100% cytoplasmic staining and 88. 9% perinuclear staining in squamous cell carcinoma. Since GST-π was significantly over-expressed in permalignant and malignant skin lesions compared with normal skin, it is suggested GST-π was induced during mouse skin malignant processing,specially GST-π in upper-layer staining and perinuclear staining can be considered as a tumor marker in skin carcinogenesis.
