摘要
目的观察人结直肠癌细胞被抗肿瘤药物作用后对不同药物的耐受情况。方法人结直肠癌LOVO细胞系分别在体外与氟尿嘧啶(5-FU)和阿霉素(ADM)作用,成功诱导LOVO/5-FU和LOVO/ADM两个耐药细胞株,体外细胞毒性实验观察它们对5-FU、丝裂霉素(MMC)、ADM、顺铂(DDP)、氨甲喋呤(MTX)、阿糖胞苷(Arac)等6种药物敏感性。结果两株细胞对5-FU和ADM均有耐药,耐药指数分别为16.48、23.17,LOVO/5-FU对MMC、ADM及DDP为3.37、2.22、2.70倍耐药,LOVO/ADM对MMC、5-FU及DDP为4.96、11.66、3.70倍的耐药。结论两株耐药细胞均产生了多药耐药(multidrugresistance,MDR),药物引起的耐药一般是交叉耐药。
Objective To investigate the resistant patterns of acquired resistance of colorectal cancer cell lines to anti cancer drugs. Methods Two resistant sublines , LOVO/5 FU and LOVO/ADM, were established with colorectal cancer LOVO cell separately selected by 5 fluorouracid (5 FU) and adriamyicn (ADM) in vitro. The susceptibility of sublines to 5 FU ,mitomycin (MMC),ADM,cisplatin (DDP),methotrexate (MTX) and arac were studied using MTT methods . Results Two resistant sublines were cross resistant to 5 FU, MMC, ADM and DDP , resistant index varied from 2.22 to 23.17. Conclusions Two sublines are all multidrug resistance (MDR), and anti cancer drug can cause cross resistant cell lines .
