细胞黏附分子与胃癌发生及转移关系的实验研究 被引量:7

Experimental study on relationship of cell adhesion molecule gene with canceration and metastasis of gastric cancer

摘要 目的探讨细胞黏附分子在胃癌发生和转移中的作用。方法运用cDNA基因芯片和逆转录聚合酶链反应(RT-PCR)技术检测25例胃癌标本神经细胞黏附分子(NCAM)、细胞间黏附分子(ICAM)、血管细胞黏附分子(VCAM)的表达。结果NCAM、ICAM及VCAM基因在25例胃癌患者中表达显著增高(Cy5/Cy3>2.0),胃癌转移组明显高于无转移组(P<0.05)和正常对照组(P<0.05),RT-PCR与基因芯片检测结果相同。结论基因芯片能够为胃癌的相关基因分析提供特异和可靠的数据,胃癌的发生和转移可能与胃组织中NCAM、ICAM、VCAM基因表达升高有关。 Objective To investigate the role of cell adhesion molecule gene in canceration and metastasis of gastric cancer. Methods The expression of neural cell adhesion molecule (NCAM) gene, intercellular adhesion molecule (ICAM) gene, and vascular cell adhesion molecule (VCAM) gene in 25 gastric carcinoma samples was detected by cDNA microarray and reverse transcriptase polymerase chain reaction (RT PCR). Results The expression of NCAM, ICAM and VCAM gene elevated in all the samples, and was significantly higher in the group with metastasis than that without metastasis (P< 0.05) and that of normal group(P< 0.01) by cDNA microarray. And same result was obtained by RT PCR.Conclusions The cDNA microarray is a powerful technique in analysis for the genes associated with gastric cancer. Upregulated expression of cell adhesion molecule genes might be related with the canceration and metastasis of gastric cancer.[
