摘要
目的 建立以查菲埃立克体P2 8融合蛋白为抗原的诊断犬埃立克体感染的免疫印迹。方法 诱导PinPointTMXa- 3 /P2 8重组质粒在Ecoli细胞内高效表达查菲埃立克体P2 8融合蛋白 ,以该重组蛋白作抗原与犬血清行免疫印迹。结果 P2 8融合蛋白与已知的 3份犬埃立克体感染血清呈阳性反应 ,与正常犬血清反应为阴性 ,在 165份来自犬埃立克体病疫区的待检犬血清中有 4 3份 ( 2 6% )与P2 8反应阳性。结论 查菲埃立克体P2 8融合蛋白可作为犬埃立克体感染的特异性诊断抗原 ,以其建立的免疫印迹可用于犬埃立克体病的诊断。
Aim To establish a method for diagnosis of Ehrlichia canis infections by immunoblot with P28 fusion protein of Ehrlichia chaffeensis .Methods P28 fusion protein of E chaffeensis was yielded in E.coli cells transformed by PinPoint TM Xa-3/P28;P28 fusion protein was used as an antigen to react with the dog sera in immunoblot.Results The positive results were observed with 3 sera from the dogs infected with E.canis and the negative results were obtained with the sera from normal dogs in immunoblot;43 of 165 serum samples (26%) collected in an endemic area of E.canis were positive in immunoblot.Conclusion P28 fusion proteins of E.chaffeensis may be used as an specific antigen for the serological diagnosis of dog ehrlichiosis,and the immunoblot established with P28 is useful to diagnose this disease.
