弓形虫表面抗原蛋白基因SAG1在骨骼肌中的表达 被引量:5

摘要 构建弓形虫速殖子表面抗原蛋白SAG1的真核表达载体 ,直接注入小鼠骨骼肌 ,观察该基因的表达程度和表达定位 ,研究利用骨骼肌异源表达SAG1,探讨发展一种简易、有效和安全的基因免疫途径的可能性。免疫组化结果显示 ,直接肌肉注射重组真核表达载体pcDAN3-SAG1可使SAG1在免疫小鼠骨骼肌中表达 。 A eukaryotic expression vector containing Toxoplasma gondii SAG1 surface antigen gene was constructed .The DNAs of the recombinant plasmid were directly injected into the skeletal muscle of mouse.The expression efficiency and the location of the expressed product were assaied to investigate the possibility of ectopic expression of the SAG1 gene in skeletal muscle,which would be benefit to develop a safe and convenient gene vaccination method for the prevention of Toxoplasmosis.Immunohistochemistry results showed that mouse skeletal muscle efficiently expressed SAG1 in vivo after direct plasmid injection,which laid a way to induce the host to generate protective anti-SAG1 antibody.
