胸腺内注射异基因脾细胞诱导移植心免疫耐受的实验研究

摘要 目的 通过大鼠同种异体心脏异位移植 ,探讨胸腺内注射异基因脾细胞对移植心的影响。方法 用供体脾细胞 (SC)在心脏移植前 2 1d预处理受体鼠 ,术后 1周每日腹腔注射环孢素A(CsA) 10mg/kg。检测移植心存活时间、混合淋巴细胞反应 (MLR)及细胞介导的细胞毒试验 (CML)。结果 胸腺注射脾细胞抗原联合短期应用CsA组的移植心平均存活时间较其他组显著延长 (P <0 .0 5 ) ,移植心耐受的受体鼠中MLR增殖活跃 ,产生免疫耐受的受体鼠的T细胞对供体鼠的靶细胞无明显的细胞毒作用。结论 胸腺内注射供体脾细胞抗原预处理受体同时联合短期应用CsA ,可诱导产生移植心脏的免疫耐受 ,且诱导移植耐受的抗原量以 2 .5× 10 Objective The experiment of cardiac allograft is to investigate the influence of transplanted heart pretreated by intranthymic injection of spleen cells in tranthymic.Method We investigated the specificity of tolerance produced by the pretreatment of spleen cells and CsA in rats model of heterotopic heart transplantation with Wistar donors 21days before cardiac transplantation.This aim is to test the time of cardiac allograft survival, mixed lymphocyte reaction (MLR) and cell-mediated cytotoxicity (CMC).Result In tranthymic injection of spleen cells combined with CsA intraperitoneally could significantly prolong graft. The Wistar cardiac allograft survival is long (average 72 days) in pretreated recipients. It demonstrated that MLR was active in long-term cardiac allograft recipients and T cells obtained from the unresponsive animals had detectable cytoxicity to donor targets.Conclusion The results confirm the tolerance induced in a cardiac allograft by intrathymic and CsA in pretreatment recipients. The quantity of alloantigens making immune tolerance was better to adapt 2.5×10 7 spleen cells.
