摘要
目的 探索用CalyculinA诱导外周血淋巴细胞染色体发生凝聚的最优化方法。方法 采用L9(34 )正交试验 ,对影响CalyculinA诱导染色体凝聚方法的 3个主要影响因素 (CalyculinA的终浓度 ,CalyculinA的处理时间 ,细胞培养时间 )进行探讨。结果 CalyculinA的终浓度 ,CalyculinA的处理时间 ,细胞培养时间不同水平的影响具有统计学意义 (P <0 .0 1)。结论 淋巴细胞培养 48h ,培养结束前 2h加入CalyculinA ,CalyculinA终浓度为5
Objective To explore the best method of PCC (premature chromosome condensation) induced by Calyculin A.Methods three main factors influcing the method of PCC induced by Calyculin A were evaluated with L 9(3 4) orthogonal design.Result It was shown by statistics analysis that there was significant diversification ( P <0.01) in both three main factors (the Calyculin A final density,the treatment time of Calyculin A,and the culture time).Conclusion The result is the best when the Calyculin A final density is 50nmol/L,the treatment time of Calyculin A 2h, and the culture time 48h.
