中药脑络通对大鼠脑缺血再灌注损伤血清与脑组织TNF-α、IL-1_β含量影响的分期观察

Periodic Observation of The Effects of Naoluotong on TNF-α ,IL-1_β Level in Serum and Brain Tissue During Cerebral Ischemic-reperfusion Damage in Rats

目的 :探讨中药脑络通改善脑缺血损伤的作用机制。方法 :多因素复合制作气虚血瘀证大鼠脑缺血动物模型 ,采用放免法测定血清与脑组织匀浆肿瘤坏死因子 (TNF α)和白细胞介素 1β(IL 1β)含量。结果 :(1)预防治疗组血清和脑匀浆TNF α、IL 1β 含量明显降低 ;(2 )急性治疗组脑匀浆TNF α 含量明显降低 ;(3)慢性治疗组血清TNF α 含量明显降低。结论 :(1)具有益气活血功效的中药脑络通不同时期的给药 ,可以不同程度降低大鼠脑缺血后病理性升高的TNF α、IL 1β 等指标 ;(2 )脑络通对于脑缺血损伤的全过程均有较好的治疗作用 ,尤其是对于发病前高危状态的预防性治疗。

Objective:To study the mechanism of NaoLuoTong in preventing and treating on cerebral ischemic reperfuson damage of experimental rats with Qi deficiency and blood stasis syndrome.Methods:The cerebral ischemic model with Qi dificiency and blood stasis syndrome was established.The level of Tumor necrosis factor alpha(TNF α)、Interleukin 1 beta(IL 1 β)in serum and brain tissue were observed.Result:(1)Prevent therapy of NaoLuoTong could remarkably decrease the level of TNF α、IL 1 β in serum and TNF α、IL 1 β in brain tissue.(2)The therapy of NaoLuoTong in acute cerebral ischemia could remarkably decrease the level of TNF α in brain tissue.(3)The therapy of NaoLuoTong in chronic cereberal ischemia could remarkably decrease the level of TNF α in serum.Conclusion:(1)The results reveal that NaoLuoTong could prevent the cerebral ischemic reperfusion damage by decreasing the pathologic high level of TNF α、IL 1 β in serum and brain tissue.(2)The above all results suggest that NaoLuoTong could be used to treat the cerebral ischemic in all times,especially for prevention.
