摘要
目的 :测定华中五味子中五味子甲素的含量。方法 :样品用环己烷超声提取 ,提取液过滤 ,蒸干 ,残渣用甲醇溶解后 ,经高效液相色谱仪测定。色谱柱 :PHENOMENEX C1 8(5 μm,4.6 mm× 2 5 0 m m)柱 ;流动相 :乙腈—甲醇—水 (15∶ 15∶ 10 ) ;检测波长 :2 5 4nm。结果 :华中五味子中五味子甲素与其它组分良好分离。五味子甲素保留时间分别约为 14.6分钟。五味子甲素对照品线性浓度范围 1.0~ 80 μg· ml- 1 ,r=0 .9996 ,平均回收率为 96 .5 3% ,RSD为1.44 %。结论 :本法灵敏度高、操作简便、结果准确 。
Objective: To determine the concentration of Deoxyschizandrin in Schisandra Sphenanthera Rehd.et Wils. by HPLC. Methods: The sample was extracted with cyc-hexane through supersonic wave. The extracted solution was filtrated and evaporated to dryness. The residue was resolved with methanol and determined by HPLC with PHENOMENEX C18(5μm, 250х4.6mmID) column, methanol-acetonitrile-water (15∶15∶10) as mobile phase. The wavelength for detection was 254nm. Result: The retention time of Deoxyschizandrin was 14.6 minutes. The standard curves of Deoxyschizandrin were linear over the concentration range of 1.0~80 μg/ml(r=0.999 6). The average recovery of Deoxyschizandrin was 96.53% and RSD was 1.44%. Conclusion: This method was sensitive, rapid and accurate for the measurement of Deoxyschizandrin in Schisandra Sphenanthera Rehd. et Wils.
