摘要
目的建立一种敏感、简便、快速的恶性疟诊断方法。方法应用薄层色谱法检测体外培养的不同虫株恶性疟原虫感染红细胞中的恶性疟原虫乳酸脱氢酶 (LDH-p)。结果该方法的敏感度可达 1 0个原虫 /μl~ 2 0个原虫 /μl血。检测感染期相同、原虫密度均为 5%的恶性疟原虫 FCC-1 /HN株、FCC-1 /AH株、FCC/YN株感染红细胞的最高阳性稀释度不同 ,提示不同虫株恶性疟原虫体内的 LDH可能存在着差异。整个检测过程3 0 min,不需特殊设备 ,费用低廉。结论薄层色谱法检测恶性疟原虫乳酸脱氢酶敏感、简便。
Objective To establish a sensitive, simple and quick method for diagnosis of falciparum malaria. Methods Thin layer chromatography was used to detect lactate dehydrogenase (LDH p) in different P. falciparum strains (all with 5% parasite density and in ring form) of FCC 1/HN, FCC 1/AH and FCC/YN cultured in vitro. Results The sensitivity of the method was 10 20 parasites/μl blood. The maximum positive dilutions were different among strains indicating that the LDH p in different strains might have discrepancy. Conclusions To detect LDH p of P. falciparum by thin layer chromatography would be a practical method for diagnosis of falciparum malaria.
