摘要
目的初步评价 CSP基因型简捷 PCR鉴定法在对间日疟原虫进行分型时的实用性。方法利用套式等位特异 PCR,检测间日疟原虫环子孢子蛋白 (CSP)基因结构多态 ,以产生的多样性 DNA片段作为对间日疟原虫分型的依据。结果经检测 4 2份被镜检确诊为间日疟滤纸血样 ,被分型 3 3份 ,分型率为 78.57% ,温带型占 3 0 .3 0 % (1 0 / 3 3 ) ,热带型占 69.70 % (2 3 / 3 3 ) ,未检到 PV- 型间日疟原虫。当第二次对 3 3份血样检测时93 .94 % (3 1 / 3 3 )被分型 ,其分型符合率为 77.4 2 % (2 4 / 3 1 )。结论该方法简便、快速 ,且分型不受采血时机限制 ,但作为云南间日疟的分型方法时 。
Objective To estimate initially the practicality of circumsporozoite protein (CSP) genotypes of Plasmodium vivax . Methods Using nest allele specific PCR to check the structure diversity of P.vivax CSP gene and typing blood samples with P.vivax according to different size band. Rssults 33 out of 42 samples were typed with a typing rate of 78.57% in which temperate zone family strain amounted to 30.30%(10/33), tropical zone family strain 69.70%(23/33) and no PV type 2 found. When the 33 samples examined for second time, the coincident rate was 93.94% (31/33) with a conformation rate of 77.42% (24/31) only. Conclusions Nest allele specific PCR is rapid, convenient to check the structure diversity of temperate zone family strain, tropical zone family strain and PV type 2 but the stablity of the test should be enhanced in future.
