摘要
从湖北省真菌传小麦黄花叶病病毒分离物中抽提病毒总 RNA,应用逆转录 -聚合酶链式反应(RT- PCR)方法 ,合成病毒外壳蛋白 (CP)基因 c DNA。对所获得的 c DNA克隆进行序列测定及分析 ,结果表明 ,湖北省真菌传小麦黄花叶病病毒分离物与小麦梭条斑花叶病毒 (WSSMV)、大麦黄花叶病毒 (Ba YMV)、大麦和性花叶病毒 (Ba MMV)等病毒 CP基因相应序列的同源性均低于 70 % ,而与报道的小麦黄花叶病毒 (WYMV)分离物 CP基因核苷酸及编码氨基酸序列同源性均超过 97%。表明湖北省真菌传小麦黄花叶病病原应为小麦黄花叶病毒 (WYMV)。
Total viral RNA was extracted from an isolate of fungal transmitted wheat mosaic virus in Hubei Province and was used to synthesize complementary DNA to the region coding for its coat protein by reverse transcription polymerase chain reaction. cDNA sequencing and analysing was carried out. Sequence comparison of coat protein genes showed that the Hubei isolate shared lower than 70% identities with WSSMV, BaYMV and BaMMV in Bymoviruses . However, the identity between the Hubei isolate and other WYMV isolates reported previously was higher than 97% both in nucleotides and amino acids. Thus, this result made it clear that the fungal transmitted wheat mosaic virus in Hubei Province is WYMV.
