摘要
用甜樱桃矮化砧木吉塞拉(Gisela)5号、7号(Prunus cerasus x P.canescens)无菌生根苗的叶片作为外植体,进行不定芽再生植株的研究。以WPM+BA 5~7 mg/L+IBA 0.1~1.0mg/L做培养基,不定芽再生率高达70%;吉塞拉5号的再生率明显高于7号;培养基中高浓度的细胞分裂素抑制吉塞拉7号的不定芽再生。不定芽的增殖、生根、温室锻炼、大田移栽均已获得成功,同时观察了植株在大田的生长状况。该成果可用于樱桃转基因育种和多倍体植株培育的研究。
Plants were regenerated from in vitro leaf segments of plantlets of Gisela 5 and Gisela 7, two cherry dwarf root- stocks on the WPM media supplemented with 6-BA 5~7 mg/L and IBA 0.1~1 .0 mg/L. High regeneration frequency was ob- tained from Gisela 5, accounted for 70%. The ability shoot organogenesis from Gisela 5 was much higher than Gisela 7. High 6-BA concentration inhibited shoot regeneration of Gisela 7. The adventitious shoots were multiplied and rooted. The plantlets were transplanted into soil and established in the field. It is believed that this study is useful for application in cherry gene transferring and genome breeding.
