小鼠诱导型环氧化酶基因敲除对脑缺血损伤的保护作用 被引量:7

摘要 目的 观察敲除诱导型环氧化酶 (cyclooxygenase ,COX2 )基因对小鼠脑缺血再灌注后脑梗死体积、前列腺素E2(prostaglandinE2 ,PGE2 )含量的影响。方法 应用COX2基因敲除小鼠 ,制备小鼠短暂性局灶脑缺血模型 ;采用TTC染色及计算机图像分析测定脑梗死体积 ;应用ELISA方法测定PGE2 含量。结果 COX2基因完全敲除鼠的脑梗死体积显著小于杂合子组和未敲除COX2基因组 ,而杂合子组脑梗死体积与未敲除COX2基因组相比无显著差异。COX2基因完全敲除鼠缺血 1h再灌注 2 4h后 ,PGE2 含量与杂合子组和未敲除COX2基因组相比显著降低 ,而杂合子组PGE2 含量与未敲除COX2基因组相比无显著差异。结论 COX2参与了脑缺血再灌注损伤 。 Objective To investigate the effect of COX2 gene knockout on cerebral infarction volume and PGE 2 content of brain after ischemia reperfusion in mice.Methods Focal ischemia was produced in mice using suture technique.TTC staining and image analysis were used to measure the infarction volume.PGE 2 content is measured by ELISA.Results COX2 gene knockout mice showed significantly smaller infarct volume as compared with the control group (P<0 01).Twenty four hours following 1h of MCA occlusion,COX2 gene knockout mice showed significantly decreased level of PGE 2 (P<0 01).Conclusion The results confirm the role of COX2 in mediating ischemic neuronal injury following temporary focal ischemia.
