摘要
目的 :从分子水平检测中药预防肝纤维化的作用及机理。方法 :利用CCl4复合因素对SD大鼠制造肝纤维化模型 ,然后用中药灌胃进行预防 ,检测其生化指标、肝纤维化指标、病理情况以及肝组织中AL RmRNA表达量。结果 :HA、PCⅢ、LN、PLD的升降与病理组织切片反映的肝纤维化程度相一致 ,在肝纤维化形成的同时给予中药预防 ,ALRmRNA的表达增强。结论 :联合检测生化、肝纤维化指标 ,可提高肝纤维化早期诊断 ;通过促进ALR的表达可延缓或减轻肝硬化的发生。
Objective:To study the mechanism of preventive effect of Chinese herbs on the CCl 4 induced hepatic fibrosis in molecular level.Method:After hepatic fibrosis model of rat was established with CCl 4 and Chinese herbs were administrated at the same time of CCl 4 injection,the level of ALRmRNA expression,biochemical and hepatic fibrosis parameters,and pathologic changes were determined.Result:The concentration of ALT,AST,HA,LN,PLD and PCⅢ in serum of hepatic fibrosis rat,and the level of ALR mRNA expression in liver of hepatic fibrosis rat determined by RT PCR increased.After Chinese herbs were administrated by mouth into rat at the same time of CCl 4 injection,the level of ALR mRNA expression in liver cells was higher but the concentration of HA,LN,PLD and PCⅢ in serum were lower than that of the group of hepatic fibrosis model.Conclusion:ALR was involved in liver repair when hepatic fibrosis occurred,and Chinese herbs could prevent hepatic fibrosis by promoting ALR gene expression.
