摘要
目的探讨大肠癌中 p16蛋白失表达及其意义 ,以及 p16蛋白与细胞增殖活性的关系。 方法应用免疫组化EnVision法检测 6 0例大肠癌和 47例大肠腺瘤中 p16蛋白及Ki- 6 7抗原的表达。 结果 1 大肠癌、大肠腺瘤中 p16失表达率分别为 2 6 .7%、2 5 .5 % ,二者无显著性差异 (P >0 .0 5 )。 2 随肿瘤分化程度的下降 ,p16蛋白的失表达率增高 ,p16表达程度明显下降 (P <0 .0 5 ) ,但 p16表达的改变与Dukes 分期、淋巴结转移等无关 (P >0 .0 5 )。3 大肠癌Ki- 6 7阳性表达率为 38.1%± 10 .1% ,较腺瘤 2 4.3%± 8.3%显著增高 (P <0 .0 1)。大肠癌低分化组Ki- 6 7阳性表达率较高、中分化组增高 (P <0 .0 1)。 4 大肠癌 p16阴性组Ki - 6 7阳性表达率 37.6 %±11.47%与 p16阳性组 38.18%± 9.6 8%相比 ,无显著差异 (P >0 .0 5 )。 结论 p16蛋白失表达与大肠癌分化呈负相关。大肠癌Ki- 6 7阳性表达率高于腺瘤 ,而且分化越差 ,Ki- 6 7表达越高。p16表达的改变与Ki- 6
Objective To evaluate the expression and significance of p16 protein in colorectal carcinoma, and to study the relationship between p16 and cellular proliferation activity. Methods The EnVision immunohistochemical method was used to detect the expression of p16 and Ki-67 in 60 cases of colorectal carcinoma and 47 cases of colorectal adenoma. Results The negative rates of p16 in colorectal carcinoma and adenoma were 26.7% and 25.5%, respectively. Ki-67 positive expression rates were higher in colorectal carcinoma than in adenoma. Both the negative rate of p16 and Ki-67 positive expression rate in poorly differentiated carcinoma were higher than in well and moderate differentiated carcinoma. There was no significant correlation between p16 and Dukesstage as well as lymph node metastasis. Also no significant correlation was found between p16 and Ki-67. Conclusion p16 protein expression in colorectal carcinoma was related to differentiation. To some extent, p16 expression may reflect malignant tendency. Both the biologic effects of p16 and the relationship between p16 and cellular proliferation may be associated with multiple factors.
