体外培养的人牙周膜细胞成骨样细胞表型特征的研究

Osteoblast-like characteristics of human periodontal ligament cells in vitro

目的 对体外培养的人牙周膜细胞成骨样细胞的表型特征进行研究探讨。方法 组织块 法培养人牙周膜细胞，放射免疫法检测条件培养基中碱性磷酸酶（ＡＬＰ）和骨钙素（ＯＣＮ）的分泌活性，免疫 组化法检测非分泌型的ＡＬＰ和ＯＣＮ的表达，原位杂交方法检测二者的ｍＲＮＡ水平的表达。结果 人牙 周膜细胞具有成骨细胞的部分表型，碱性磷酸酶在蛋白水平包括分泌和非分泌型及基因转录水平都有一 定的表达，随培养时间的变化不大；而不具有成熟的成骨细胞的表型特征——骨钙素的表达，其无论是在 基因和蛋白水平都没有表达二这一结果为进一步研究人牙周膜细胞在机械力作用下参与骨改建打下基础。

Objective To study the osteoblast-like characteristics expressed he human periodontal ligament cells in vitro. Method Human periodontal ligament cells (hPDLC) were cultured by explant method. RIA examina-tion indicated the expression of secreting ALP and OCN in conditional media, histoimmunochemistry determined the non-secreting ALP and OCN in cells, and hybridization in situ examined the expression of above two proteins in mRNA level. Results Human periodontal ligament cells express ALP both in protein level( include secreting and non-secret-ing) and in mRNA level, while no expression of OCN - as a marker of differentiated osteoblast-like cells. This suggest-ed that IIPDLC are the potential osteoblast-like cell during differentiation.
