体内抑制一氧化氮合成酶对小鼠卵母细胞成熟和排卵的影响 被引量:8

摘要 实验研究了一氧化氮合成酶抑制剂 L - NAME对昆明白小鼠卵母细胞成熟和排卵的影响。初情期前的昆明白小鼠注射 PMSG促进发情 ,注射 h CG促进排卵 ,并在注射 h CG前后各 3h注射 L- NAME。注射h CG后 18h颈椎脱臼处死小鼠 ,取输卵管冲卵。统计排卵数及卵母细胞的减数分裂情况发现 ,注射 L- NAME后排卵数明显减少 ,卵母细胞减数分裂恢复异常 ,大部分排出的卵母细胞处于 MI期 ;PB1的排出率降低 ,形态异常或退化死亡的卵母细胞数增加。结果直接证明 :在体内 ,NO可促进小鼠卵母细胞的减数分裂的恢复 ;NO对于小鼠排卵具有重要的促进作用 ;NO对小鼠卵母细胞质的成熟有重要影响 ;抑制 NO合成后 。 To examine the effect of inhibiting nitric oxide synthase(NOS) on the number of ovulated oocytes and on the oocytes meiotic maturation, the immature Kunming white mice were super ovulated with PMSG and hCG. Three hours before and 3 hours after hCG injection, the mice were treated with 1 mmol L NAME. The mice were killed 18 hours after hCG injection by cervical dislocation, and the oocytes present in the oviduct were flushed out, counted and classified for stages of meiosis. The ovulated oocytes obstained from mouse treated with L NAME were significantly fewer than that of the control and more oocytes displayed the abnormal meiosis stage, the most oocytes were at metaphase Ⅰ stage. The results indicate that ovarian nitric oxide is required for maximal ovulation and oocyte meiotic maturation in mouse.
