摘要
目的 探讨 EB病毒编码小 RNA (EBERs)多态性与鼻咽癌发病趋势的联系。方法 巢式 PCR扩增 E-BER- 1和 EBER- 2序列 ,直接测序 ,比较鼻咽癌高发区广东地区鼻咽癌组织、正常人鼻咽黏膜组织和鼻咽癌低发区哈尔滨地区正常人漱口液感染的 EB病毒 EBERs序列差异。结果 广东地区鼻咽癌组织感染的 EBERs具备 1型病毒的特征。与原型株 B95 .8相比 ,广东地区鼻咽癌组织来源的 EBER- 2基因存在 8处碱基替换和 2处碱基缺失。但是 ,广东正常人鼻咽黏膜组织和哈尔滨地区正常人漱口液来源的 EBERs也有相同的碱基替换和缺失。结论 鼻咽癌高发区和低发区流行的 EB病毒 EBERs序列呈现高度一致性。
Objective To analyze the DNA sequence of Epstein-Barr virus-encoded small RNAs (EBERs) and to detect possible polymorphism associated with nasopharyngeal carcinoma (NPC).Method EBERs fragments were amplified with nest PCR method from NPC tissue, normal nasopharynx in Guangdong area and throat washings from normal donors in Haerbin area.PCR productions were subjected to sequence and then analyzed with BLAST software.Results EBERs from NPC tissue had the basic characteristics of type 1 EBV and also showed 8 base pairs of substitution and 2 base pairs of deletion in EBER-2. The sequences of EBERs from normal nasopharynx in Guangdong and from throat washings in Haerbin were identical to that from NPC tissue in Guangdong.Conclusion The EBERs sequences derived from endemic and non-endemic area of NPC are highly homogeneous.\;
