人参总皂甙诱导人内皮细胞表达造血生长因子的实验研究 被引量:2

摘要 目的:研究和论证人参促进血细胞发生及“补气生血”的现代生物学机理。方法:采用造血生长因子生物活性检测与免疫细胞化学等技术,研究经人参总皂甙(TSPG)诱导的人脉静脉内皮细胞株(Ecv304)上清液(HEcCM)中造血生长因子的活性及GM-CSF、IL-3和IL-6在内皮细胞的表达。结果:TSPG体外诱导制备的HEcCM能显著促进CFU-GM和CFU-Mix的增殖;同时,TSPG能不同程度地促进内皮细胞表达GM-CSF、IL-3和IL-6蛋白。结论:本研究提示TSPG能上调造血微环境中的内皮细胞分泌造血生长因子来调节血细胞的生长,这或许是人参“补气生血”的现代生物学机理之一。 Objective:To study modern biological mechanism for hematonic of Ginseng.Methods:By using the techniques of biological assay of hematopoietic growth factor and immunocytochemistry, the activity of hematopoietic growth factors of the conditioned culture media of human umbilical vein endothelial cell (Ecv304) (HEcCM) and the expression of GM-CSF,IL-3,IL-6 in the endothelial cell induced by Total Saponins of Panax Ginseng (TSPG) were studied .Results:HEcCM prepared with TSPG markedly promoted the proliferation and differentiation of CFU-GM and CFU-Mix. The expression of GM-CSF,IL-3 and IL-6 in the endothelial cell induced by TSPG was much stronger than that of the control.Conclusion :We speculate that TSPG augmented endothelial cell capacity to support hematopoiesis,and this property might result from the upregulation of hematopoietic growth factors expression.
