摘要
目的:观察牛磺酸对白细胞介素-1β(1L-1β)、肿瘤坏死因子(TNF)和γ-干扰素(IFN-γ)诱导的胰岛细胞凋亡的影响。方法:应用体外单层培养的Wistar大鼠胰岛细胞,分别检测IL-1β、TNF和IFN-γ以及牛磺酸对胰岛细胞凋亡细胞百分率,培养液中NO含量及NOS活性以及DNA片段的影响。结果:IL-Iβ、TNF和IFN-γ联合可诱导胰岛细胞凋亡率及DNA片段明显增加,同时NO含量和NOS活性亦明显升高(P<0.01);牛磺酸能阻断上述细胞因子的作用(P<0.01),且有剂量依赖性。结论:NO可能是介导上述细胞因子引起胰岛细胞凋亡的重要途径之一。牛磺酸能够改善细胞因子诱导的胰岛细胞凋亡,其机制可能与抑制NOS活性从而减少NO的生成有关。
Objective: To observe the role of taurine on apoptosis of isolated pancreatic islet cells induced by combination of IL-1β,TNF and IFN-γ.Methods: Isolated pancreatic islet cells from Wistar rat were incubated in monolayer in vitro.NO production,NOS activity ,apoptotic percentage of islet cells and DNA fragmentation in pancreatic islet cells incubated with combination of IL-1β,TNF and IFN-γ, taurine were measured .Results:Combination of IL-1β,TNF and IFN-γ could induce a significant increase in apoptotic percentage of pancreatic islet cells and DNA fragmentation.NO production and NOS activity also markedly increased (P<0.01),which were blocked by addition of taurine to the pancreatic islet cells (P<0.01)。Conclusion:NO might be one of the important ways of inducing apoptotic cell death of pancreatic islet cells by a combination of IL-1β,TNF and IFN-γ.Taurine attenuates the cytokines-induced β cells apoptosis, the mechanism of which may be the inhibition of NOS and the decreasing of NO production.
