摘要 目的:建立反相高效液相色谱分析方法测定人血清中左旋多巴的浓度。方法:血样经6%的高氯酸沉淀蛋白,以纯水为流动相,紫外280 nm处检测。结果:在0.84~26.88 μg·ml-1浓度范围内,峰面积与浓度呈良好的线性关系, r=0.9993,最低检测血清浓度为0.84 μg·ml-1,平均回收率为(100. 0±4.3)%,高中低3个浓度样品的日内和日间测定相对标准偏差RSD均小于5%。结论:本方法可用于左旋多巴的临床药动学研究及临床血药浓度监测。 OBJECTIVE To establish an RP-HPLC method for determining serum level of levadop a. METHODS The samples were precipitated with 6% perchloric acid.The mobile phase was pure water.The detector was operated at 280 nm. RESULTS A better linearity was obtained from 0.84~26.88 μg of levodopa per ml for serum with a good correlation coefficient ( r =0.9993 ).The limit of detection was 0.84 μg·ml-1 for serum.The ave rage recovery was (100.0±4.3)% ,and the RSD within-day and b etween-day were less than 5%,respectively. CONCLUSIONS The method is simple,precise and usable for pharmacokinetic studies of levodopa in human.
