血管内皮生长因子反义RNA对体外培养的人脑垂体瘤细胞增殖和凋亡的影响

The effect of VEGF antisense RNA on the proliferation activity and apoptosis of human pituitary adenoma cells in vitro

目的 :研究血管内皮生长因子 (VEGF)反义RNA对体外培养人脑垂体瘤细胞增殖和凋亡的影响。方法 :将VEGF反义cDNA(反义组 )、正义VECFcDNA质粒 (正义组 )转染垂体瘤细胞 ;应用免疫组化法和TUNEL法分别检测肿瘤细胞增殖指数及细胞凋亡。结果 :反义组细胞增殖指数及细胞凋亡数量与正义组、未转染组相似 ,差异无显著性 (P <0 0 5)。结论 :VEGF反义RNA对体外培养人脑垂体瘤细胞增殖和凋亡无明显影响 。

Objective:To study the effect of VEGF antisense RNA on the proliferation activity and apoptosis of human pituitary adenoma cells in vitro.Methods:Carrying with VEGF antisense cDNA,sense VEGF cDNA plasmid were transfected into pituitary adenoma cells,the proliferation activity and apoptosis were detected by immunohistochemistry and TUNEL method.Results:There was no significant difference of the proliferation activity and apoptosis between the antisense group,sense group and control group (P>0 05).Conclusion:VEGF had no effect on the cell proliferation and induction of cell apoptosis,and provided evidence for gene therapy of pituitary adenoma.
