摘要
报道了青天葵叶片离体培养的研究结果 ,试验表明 :青天葵叶片在MS附加BA 0 .3mg·L- 1+ 2 ,4 D 1mg·L- 1的培养基上可产生俞伤组织 ;将愈伤组织转入附加BA和NAA的KC培养基中可产生原球茎 ;原球茎在KC附加BA 2 .5mg·L- 1与NAA 0 .2mg·L- 1的培养基上可形成走茎 ;将走茎转入附加NAA 3mg·L- 1的H培养基中 ,可形成大量新的球茎 ;新的球茎转入KC附加w =0 .1%的活性炭和 φ =10 %的椰子水的培养基中 ,可形成完整植株 ;试管苗移栽在椰糠上成活率可达 83.3% .
The research results of Nervilia Fordii leaves in vitro culture are presented in this paper.Experiments show that: Nervilia fordii Leaves can produce callus in MS medium with BA 0.3 mg·L -1 + 2,4 D 1.0 mg·L -1 ;when transplanted in KC medium with BA and NAA,the callus can form pre corm; in KC medium with BA 2.5 mg·L -1 +NAA 0.2 mg·L -1 ,the pre corm can form amphitropous stem; in H medium with NAA 3 mg·L -1 ,the amphitropous stem can produce great amount of new corms; in KC medium with w =0.1% activated carbon+ φ =10% coconut water,new corms can become plants; when the tube plants are transplanted into coconut husk,the survive rate of tube plants can reach 83.3%.
