摘要 目的:研究60Co γ射线对Lewis肺癌细胞Fas基因蛋白表达的影响及其诱发细胞凋亡的分子机制。方法:采用免疫电镜技术,对受照Lewis细胞的小鼠移植瘤组织进行Fas基因蛋白的超微结构定位观察。结果:受照Lewis肺癌细胞的小鼠移植瘤组织,癌细胞呈Fas基因蛋白阳性表达,阳性产物分布于细胞膜表面;而未受照射的对照组Lewis细胞的移植瘤组织,癌细胞Fas蛋白全部阴性。结论:γ射线诱发Lewis肺癌细胞发生凋亡主要依赖于Fas蛋白表达的上调及Fas介导的凋亡信号传导途径的激活。 ve:To research the influence of 60Co γ- ray on Fas gene protein expression in Lewis lung cell line , and the molecular mechanism of Lewis cell apoptosis induced by radiation. Method: Immunoelectron microscopy was used to observe the expression of Fas gene protein in mouse transplanted - tumors of Lewis cell line. Results: In mouse transplanted - tumors of radiated -Lewis cell line, the cancer cells were positive for Fas gene protein which located on the surface of cell membrane. But in mouse transplanted - tumors of non - radiated - Lewis cell line, the cancer cells showed negative for Fas protein. Conclusion: Lewis cell apoptosis induced by γ - ray depend mainly on the upregulation of Fas gene protein expression and the cativation of apoptosis signaling pathway mediated by Fas gene protein.
