转VEGF基因对血管损伤后内皮修复及内皮功能的影响 被引量:5

摘要 目的 观察局部转染VEGF基因对血管球囊拉伤后内皮修复及内皮功能的影响.方法 90只新西兰大白兔随机分为3组,Ⅰ组单纯拉右髂动脉;Ⅱ组拉伤后局部转染真核表达质粒Adtrack CMV;Ⅲ组拉伤后局部转染pAdtrack CMV-VEGF 165;每组按实验终点随机分为5个亚组,术前1周开始予高脂饮食至实验终点.喂养至4周的动物处死前20min经耳缘静脉注射5%伊文氏蓝1 mg/kg,取拉伤段髂动脉用于进一步检测.结果 三组动物血脂水平保持在正常的5~10倍,组间血管损伤程度评分相似(P>0.05).Ⅲ组损伤内皮修复面积明显大于Ⅰ组和Ⅱ组(分别为97.43±8.51 vs 56.89±6.74和61.23±4 95,P<0.01),内皮依赖性舒张功能明显强于Ⅰ组和Ⅱ组,ec NOS mRNA表达水平明显高于Ⅰ组和Ⅱ组;扫描电镜显示Ⅲ组血管内皮细胞形状规则,接近正常形态,少见血细胞聚积和微血栓形成.结论 局部转染pAdtrack CMV-VEGF 165可有效促进血管损伤后内皮修复和增进内皮功能. Abstract: Objective To investigate the effectof phVEGF 165 transfer on neointima repairing and en-dothelial function after vascular balloon injury. Methods 90 New Zealand white rabbits were divided into 3groups randomly: group I (balloon injury group), groupII(pAdtrackCMV group) and group III (pAdtrack CMV-VEGF 165 group). All animals had been given hyperc-holesterol diet for 7 days before experiment and continuedto receive hypercholesterol diet until killed. Each groupwere divided into five subgroups according to the sacrificetime. Blood samples and iliac arteries were harvested forfurther analysis. Results In group III, the degree ofreendothelialization was higher and the endothelium-de-pendent relaxations to Ach and pretreated with L-Arg+Ach were significantly improved than in group I and II(P<0.01), and a marked production of ecNOS werefound from 1 week to 4 week time. Scanning electron mi-croscopy showed that the endothelial cells were almostnormal and aligned in the direction of blood flow in groupIII.Conclusion Local phAdtrackCMV-VEGF165 trans-fer could accelerate neointima repairing and significantlyimprove endothelial function after vascular injury.
