摘要
目的 :用体外实验方法探讨人肝癌细胞株HepG2和BEL - 740 2的放射敏感性。方法 :人肝癌细胞HepG2和BEL - 740 2体外培养 ,应用集落形成法测定 6MVX线不同剂量照射后细胞的存活率 ,计算细胞放射敏感性参数 ;应用HE染色和流式细胞术测定细胞受 5Gy照射后细胞凋亡率。结果 :HepG2的D0 为1.5Gy ,Dq值为 0 .8Gy ,n为 1.8,SF2 为 0 .45± 0 .0 5 ;BEL - 740 2的D0 为 1.6Gy ,Dq为 1.3Gy ,n为 2 .4,SF2 为0 .6 3± 0 .0 5。两株细胞受照后出现凋亡现象 ,照射后 2 4h内随时间延长细胞凋亡率逐步升高。结论 :HepG2和BEL - 740 2具有较高的放射敏感性 ,可能与其受照后发生凋亡有关。
Objective:To evaluate the radiosensitivity of hepatocarcinoma cell lines HepG2 and BEL-7402.Methods:Using cell culture of two human being hepatocarcinoma cell lines HepG2 AND bel-7402,we measured their surving fraction after being irradiated by 6MV X-ray with doses from 0~10Gy by means of clonogenic assay.Radiosensitivty indexes have been calculated.We also determined their apoptotic rates after being irradiated by 5Gy X ray.Results:The D 0 of HepG2 is 1.5Gy,Dq 0.8Gy,n 1.8,SF 2 0.45±0.05 and the D 0 of 7402 1.6,Dq1.3,n 2.4,SF 2 0.63±0.05Gy,HepG2 and BEL-7402 showed obvious changes of apoptosis and their apoptotic rates had been increased by time after being irradiated.Conclusions:The radiosensitivity of HepG2 and BEL-7402 is rather high and it might be related to their apoptosis induced by X ray.
