丙型肝炎病毒非结构区NS5A研究进展 被引量:1

摘要 0引言 目前全球各国丙型肝炎病毒(hepatitis C virus,HCV)感染率为0.5%~4%,平均为3%左右[1-12],其中20%左右是急性肝炎,70%~80%转变成慢性肝炎,而慢性丙型肝炎一般经过20 a~30 a发展为肝硬变或肝细胞癌(HCC),HCV相关性终末期肝病已成为欧美一些国家进行肝移植的主要适应证[12-28],危害极大.HCV是正单股RNA病毒,包括5′非编码区(5′non-coderegion,5′-NCR),一个长约9400bp的单一开放阅读框(openreading frame,ORF)和3′非编码区(3′non-encode region,3′-NCR),5′-NCR其间有内源性核糖体进入位点(internal ribosome entry site,IRES)其中OFR包括结构区(structural region)和非结构区(nonstructural region),在结构区中有C,E1,E2,P7区,在非结构区中有NS2,NS3,NS4A,NS5A和NS5B区等[24,29-33].近年来,有关HCV NS5A区的结构与功能倍受关注,现就HCV NS5A作一综述.
