McAb捕获抗原检测EDS76病毒抗体的ELISA方法的建立和应用

Establishment and Application of McAb Capture ELISA for Detecting Antibodies to EDS76

将三株EDS76病毒单克隆抗体IgG混合,包被ELISA板,捕获病毒抗原后用于检测鸡血清中的EDS76抗体,以OD490nm>0.3,P/N>2.1判为阳性结果。经测定,待检血清做1∶100倍稀释时可以很好地区分阴阳性。所建方法与鸡的其它传染病血清无交叉反应,可被免抗EDS76病毒高免血清竞争性抑制。对11个鸡场330份未接种EDS76疫苗的产蛋鸡血清检测结果表明,血清阳性率最高可达93.33%。

McAb capture ELISA for detecting EDS76antibody of chicken serum was established by coating plate with3strains McAb IgG to capture EDS76virus.The result showed that optimal dilution titer of serum was1:100.As OD490nm>0.3,P/N>2.1,the result was positive.The samples can be detected specifically without cross-reaction to other infectious diseases.The positive reaction can be inhibited competitively by positive serum to EDS76from rabbit.330chicken samples from11chicken farms were detected and the positive rate of EDS76antibody was up to93.33%.
