摘要 采用正交试验 ,对宇佐美曲霉酸性蛋白酶摇瓶发酵工艺条件进行了优化。最佳发酵培养基的组成为 :玉米粉 1.5g ,豆饼粉 6 .0g ,蛋白胨 0 .7g ,氯化铵 1.0g ,氯化钙 0 .5g,磷酸氢二钠 0 .4g ;最优培养条件是 :pH值为 5 .5 ,培养温度为 32℃ ,摇床转速为 2 2 0r/min。采用优化工艺 ,酸性蛋白酶的酶活由 72 0 0U/ml左右提高到 840 0U/ml。 The fermentation process of the aspergillus usaamii 9169 which produced acidic protease was presented. The experimental results showed that the optimum culture medium was a mixture containing 1.5 g corn meal, 6.0 g soybean meal, 0.7 g peptone, 1.0 g NH 4Cl, 0.5 g CaCl and 0.4 g Na 2HPO 4. The initial pH value of the culture medium was controlled to be about 5.5. Spores of aspergilluss usaamii 9169 were inoculated into a 500 ml shake flask containing 50 ml medium. The spores were cultured at 32 ℃ for 80 h on a 220 r/min shaking table. With optimized technology adopted, the enzymic activity of the acidic protease increased to 8 400 U/ml from 7 200 U/ml.
