摘要
目的 应用单细胞电泳技术探讨黄绿青霉素 (CIT)对细胞 DNA损伤的作用 ,探索 CIT暴露的生物标志。方法 SGC-790 1细胞常规培养 ,荧光显微镜观察 CIT不同剂量对 DNA的效应。用 Kinetic彗星图像分析软件 ,对 DNA的尾长和尾长积差进行分析。结果 CIT可致细胞 DNA损伤 ;随着 CIT浓度的增加 ,DNA的迁移长度和尾长积差都明显增加 ,呈显著剂量效应关系 ,以尾长积差指标更灵敏。结论 单细胞电泳技术检测的 DNA损伤有望成为
Objective We study cellular DNA damage caused by citreoviridin using single cell gel electrophoresis technique in order to find an ideal biomarker of citreoviridin for epidemiological observation.Methods Cell culture in vitro and single cell gel electrophoresis assay.Results Cells DNA damage caused by citreoviridin has been observed in this paper. Both DNA Tail Length and Extent Tail Moment increase with the increasing of citreoviridin dose and have obvious dose dependent relation. Extent Tail Moment is more sensitive than Tail Length in DNA damage assay.Conclusions Citreoviridin has the effect on cellular DNA damage and it is possible to make an ideal biomarker for epidemiological observation.
