摘要
目的 分析柯萨奇 B3病毒 (CVB3 )中国分离株编码主要中和抗原的 VP1基因序列。方法 用逆转录 PCR技术扩增 CVB3中国分离株的 VP1基因 ,通过 TA克隆法构建 p CR2 .1-VP1并进行核苷酸序列的测定分析。结果 测序发现 CVB3中国分离株与 CVB3 Nancy株 VP1基因均编码 2 93个氨基酸 ,二者存在 5 9个核苷酸差异 ,其中 10处影响到氨基酸的编码 ,其余均为同义变异。结论 CVB3中国分离株与 Nancy株 VP1基因序列存在一定的差异 。
Objective To identify the sequence of VP1 gene which encodes the main neutralizing antigen of Coxsackievirus B3 Chinese strain.Methods The VP1 gene of CVB3 was amplified by reverse transcript polymerase chain reaction (RT PCR) and pCR2.1 VP1 was constructed by TA cloning strategy.The recombinant plasmid had been analyzed by DNA sequencing.Results Although the VP1 gene of CVB3 Chinese strain encoded 293 amino acids that was similar to the VP1 gene of CVB3 Nancy strain, there were 59 differences between them. Most of the variations were nonsense mutations. Only 10 amino acids changed due to the variations.Conclusions There were differences between VP1 gene of Chinese strain and Nancy strain, our results may be helpful to the research of immunological characteristics of CVB3 Chinese strain.
