摘要
目的 观察高浓度钙对氟细胞毒性的影响。方法 体外培养的人牙乳头间充质细胞给予不同浓度氟 10~ 2 63 0 μmol/L处理 ,将有明显氟细胞毒性的实验组同时予高浓度钙 (4 .1,10 mmol/L)处理 ,用 S四唑盐(MTT)比色法和透射电镜观察细胞活性和超微结构。结果 2 63 ,13 15 ,2 63 0× 10 -6mol/L 氟对人牙乳头间充质细胞毒性明显 ,10× 10 -3 mol/L和 4.1× 10 -3 mol/L钙对 13 15× 10 -6mol/L浓度以下氟毒性有明显拮抗作用 ,对2 63 0× 10 -6mol/L氟毒性作用不确定。
Objective Whether the interference by fluoride (F) with human dental papilla mesenchymal cell(DPMC) could be altered by altering the levels of calcium(Ca) in the medium was investigated.Methods Human DPMCs were exposed to 10~2630×10 6 mol/L(0.2 50×10 6 g/L) of F in vitro for 6~72h in the presence of high levels(4.1,10×10 3 mol/L)of Ca. The activity and microstructure of the cells were examined by MTT colorimetric analysis and TE mol/L respectively.Results In the presence of 263,1315,2360×10 6 mol/L of F the activity of human DPMCs was inhibited and the microstructure was changed. 4.1×10 3 mol/Land 10×10 6 mol/L levels of Ca seemed to modify the F effect, whereas the modification was not obvious when F content was 2 630 ×10 6 mol/L.Conclusions The effects of F could be partially antagonized by high Ca.
