摘要
目的 探讨脂质过氧化反应在兔坐骨神经火器伤后腰段脊髓细胞凋亡中的意义。方法 火器伤组靶点为兔右后肢外侧坐骨神经体表投影线中点 ;切割伤组在同一致伤水平切断坐骨神经。用DNA电泳、TUNEL染色技术对腰段脊髓细胞进行凋亡定性检测 ,用流式细胞术进行定量检测。同时测腰段脊髓超氧化物歧化酶 (SOD)活性和丙二醛 (MDA)含量。结果 火器伤组 1周和 2周时 ,腰段脊髓神经元和胶质细胞发生凋亡 ,切割伤组仅在 4周时有少量运动神经元发生凋亡。火器伤组 1d时 ,MDA含量显著下降 (P <0 0 1 ) ,3d、1周、2周时 ,MDA含量显著升高 (P <0 0 1 ) ,切割伤组MDA含量变化不明显。结论 火器伤组细胞凋亡发生时间早、数量多 。
Objective To explore the significance of lipid peroxidation in the cell apoptosis of the lumbar spinal cord of the rabbit after sciatic nerve injured by firearm.Methods In firearm injury group,the target was the midpoint of the lateral body surface projection of the right sciatic nerve.The sciatic nerve was cut at the same level in the cut group.The apoptosis regulations of motoneuron and glial cell were investigated with the technology of DNA gel electrophoresis?flow cytometry and TUNEL staining.The content of MDA and activity of SOD of the lumbar spinal cord were detected also.Results In firearm injury group,the apoptosis of the motoneurons and glial cells were confirmed by TUNEL staining?gel electrophoresis and flow cytometry at 1 week and 2 week,the content of MDA increased at 3 d?1 w?2 w( P <0 01).Only a small quantity motoneurons were positive by TUNEL staining in cut group at 4 w,and the content of MDA had not changes.Conclusions In firearm injury group,cell apoptosis occurred earlier,and the number of apoptotic cells was more than that of the cut group.Lipid peroxidation had relationship with the occurrence of cell apoptosis in firearm injury group.
