压电石英晶体微阵列免疫传感器检测hCG的实验研究 被引量:13

摘要 目的 构建一种新型压电石英晶体人绒毛膜促性腺激素 (hCG)免疫微阵列传感器。方法 抗hCG β单克隆抗体采用蛋白A(SPA)法固定在 2× 5的AT切向、基频 1 0MHz的石英晶体微阵列金膜电极表面制成抗体敏感膜 ,构成压电石英晶体hCG免疫微阵列传感器。结果 构建的hCG压电石英晶体微阵列免疫传感器对hCG的响应特性良好 ,其检测线性范围为 2 .5~ 2 50mIU ml,TSH、FSH、LH对hCG的检测基本无干扰 ;60例临床标本检测结果与荧光免疫检测法符合 (P >0 .0 5) ,两种方法的相关系数为 0 .92。结论 研制的压电石英晶体hCG免疫微阵列传感器具有灵敏度高、特异性好、成本低廉、省时、操作简单、不需标记、能在线实时检测等优点 ,对比实验表明其检测结果准确 。 Objective To construct a new type of piezoelectric quartz crystal micro array immunosensor for the determination of human chorionic gonadotropin(hCG). Methods The crystal was 10 MHz AT cut quartz with gold coated electrodes. The immobilization of the monoclonal antibody of hCG β onto the surfaces of the gold electrodes was accomplished via protein A. Results Experimental results showed that the piezoelectric immunosensor had good response to hCG,the frequency shifts were linearly dependent on the hCG concentration in the range of 2.5~250 mIU/ml,other hormones such as TSH,LH and FSH, did not interfere in the sensor's frequency. The analytical results from this approach were in satisfactory agreement with those of by fluorescent immunoassay ( P >0.05) with correlation coefficiency of 0.92. Conclusion Piezoelectric micro array immunosensor for determination of hCG is of high sensitivity,good specificity,simple operation,rapid analysis,low cost and real time detection etc. It is suitable for detecting hCG in serum and urine in clinical laboratory.
