骨质疏松大鼠胰岛素样生长因子-1血清水平和转化生长因子β_1骨表达的实验研究 被引量:7

摘要 观察骨质疏松大鼠不同时期骨密度 (BMD)、血清胰岛素样生长因子 - 1(IGF - 1)的浓度以及骨组织中转化生长因子 β1(TGF - β1)的表达 ,进一步探讨绝经后骨质疏松与细胞因子的相关性。方法 :将 6 0只10~ 12月龄的SD大鼠随机分成实验组和对照组 ,用摘除双侧卵巢的方法制备实验大鼠骨质疏松模型 ,实验组行摘除双侧卵巢术 ,对照组行假手术 ,仅切除卵巢周围的脂肪组织 ,应用HOLOGIC第 4代双能X线4 5 0 0W骨密度仪测量去卵巢不同时期 (术后 8,12 ,16周 )大鼠全身骨密度和腰椎感兴趣区域 (L1~L4)骨密度 ,并采用ELISA法和免疫组化的方法 ,检测去卵巢大鼠不同时期血清IGF - 1水平和骨组织中TGF - β1表达。结果 :全身骨密度 :去卵巢组大鼠在术后第 8周和第 12周 ,全身骨密度与对照组 (假手术组 )无明显差异(P >0 .0 5 ) ,在术后第 16周 ,开始出现全身骨密度降低 ,与对照组相比 ,有显著差异 (P <0 .0 1)。腰椎 (L1~L4)骨密度 :从术后第 8周~第 16周去卵巢腰椎骨密度均明显低于对照组 (P <0 .0 1)。血清IGF - 1浓度 :从术后第 8周~第 16周 ,去卵巢组血清IGF - 1浓度值均低于对照组 (P <0 .0 5 )。骨组织TGF - β1表达 :去卵巢组在术后第 8周骨组织中软骨细胞、成骨细胞、骨基质TGF - β1的表达程度与对照级有? Objective:To observe the bone mineral density (B MD), serum Insulin-Like growth factor-1 (IGF-1) levels and bone Transforming gro wth factor beta-1 (TGF-β 1) expression in different period of osteoporosis r at and to research relationship between postmenopausal osteoporosis and the gr owth facto r .Methods:Sixty 12-month old female SD rats were randomized into t wo groups of 30 each, the experiment group and the control group.Experimental model of osteoporosis was established by removing twin ovary. BMD of global and the l umbar spine was measured with HOLOGIC dual energy X-ra y bone densitometer in rat.Serum IGF-1 levels and bone TGF-β 1 expressions in different period of osteoporosis rat were investigated by means of ELISA and imm unohistochemitry.Results:BMD of global in the experiment group was compared with t hat in the control group, there was no significant difference ( P >0.05) at 8-12 weeks after the operation.But at 16th week, BMD of the lumbar spine in the exp eriment group was lower than that in the control group ( P <0.01).BMD of glob a l in the experiment group was lower than that in the control group( P <0.01) during different periods. From 8th to 16th week, serum IGF-1 concentrati on in the ex perment group was lower than that in the control group ( P <0.05).Chondrocyte and osteoblast showed different expressions of TGF-β 1 at the 8th week. Conclusion:TGF-β 1 and IGF-1 play a critical role in the po stmentopausal osteoporosis.
