摘要
目的 了解AML -M2a患者外周血中TCRVβ亚家族T细胞的分布和克隆性情况 .方法 利用RT -PCR扩增 14例AML -M2a患者外周血单个核细胞中 2 4个TCRVβ亚家族基因的CDR3,了解TCRVβ亚家族T细胞的分布 .阳性产物进一步经基因扫描分析了解其克隆性 .结果 14例AML -M2a患者选择性表达 3~ 10个Vβ亚家族 ,其中部分Vβ亚家族呈寡克隆增殖 .结论 AML -M2a患者TCRVβ亚家族T细胞的倾斜性分布和克隆性增殖 ,可能是机体对M2a细胞刺激所产生的特异性免疫反应 .
Objective The distribution and clonality of TCR Vβ subfamily T cells were analyzed in patients with M 2a subtype of acute myelogenous leukemia (AML-M 2a ).Methods The complementarity determining region 3 (CDR3) of TCR Vβ 24 subfamily genes in peripheral blood mononuclear cells from 14 cases with AML-M 2a were amplified using RT-PCR.The positive products were further analyzed to identify the clonality of T cells by genescan.Results Only 3~10 Vβ subfamily T cells were selectively expressed on patients with AML-M 2a .Oligoclonal expansion T cells could be found in some Vβ subfamily T cells from all 14 cases.Conclusions The skew distribution and clonal expansion of TCR Vβ subfamily T cells detected in the patients with AML-M 2a may be a rusult of the host specific immune response to tumor antigens expressed on M 2a malignant cells.
